Retinol is one of the most studied anti-aging ingredients in skincare (and for good reason!).

It's a form of vitamin A that your skin converts into Retinoic Acid, the active molecule that actually does the work in your cells.

That conversion happens in two steps: your skin first turns Retinol into Retinaldehyde (also called Retinal), then turns Retinaldehyde into Retinoic Acid.

Retinol is converted to biologically active retinoic acid via retinaldehyde by dehydrogenases in a two-step oxidation process.

Each step is a little "upgrade" toward the active form which is part of why Retinol is gentler than prescription Retinoic Acid; your skin does the work gradually. This also explains where Retinol sits in the retinoid family.

Here is the retinoid family ranked roughly by strength: Retinyl Esters (like Retinyl Palmitate) < Retinol < Retinaldehyde < Retinoic Acid.

Retinoid activity increases in that order, while tolerance runs in reverse; retinyl esters are the gentlest and retinoic acid the most irritating.

The more conversion steps an ingredient needs, the gentler (and slower) it tends to be, so Retinol lands in a nice middle spot. It's more effective than the esters, gentler than prescription options.

Once it becomes Retinoic Acid, it binds to receptors inside your cells' nuclei (called RARs and RXRs). These receptor pairs bind to specific DNA motifs called retinoic acid response elements and act like switches that turn certain genes on or off.

In practice, this means a few things happen in a formula. It:

That last two are worth a closer look.

A study that tested Retinol directly (not just prescription Retinoic Acid) found that four weeks of retinol thickened the epidermis and switched on the genes for Collagen I and Collagen III, with more procollagen I and III showing up in the skin. And after twelve weeks, facial wrinkles were visibly reduced.

Retinoids more broadly stimulate the skin's synthesis of hyaluronan and other glycosaminoglycans, part of what gives skin a plumper, more hydrated look over time.

So even the gentler OTC form is doing real structural work (not just sitting on the surface).

It's also worth knowing Retinol isn't only a wrinkle ingredient; it can help with uneven tone, dark spots, rough texture, and the look of pores as well because it speeds up turnover and influences pigment.

The research backs this up as well.

A pooled analysis of six clinical studies found that 0.1% stabilized retinol improved all signs of photoaging versus vehicle as early as week 4 and through 12 weeks, with only a few mild cases of irritation.

Another study comparing concentrations found that 0.3% and 1% Retinol were similarly effective at remodeling photodamaged skin, but 0.3% caused fewer adverse reactions when used daily (a useful reminder that more isn't always better).

Retinol is about tenfold less potent than Retinoic Acid. This is why it works as a gentler, non-prescription option that builds results over time.

Typical concentrations range from 0.1-1%, with 0.1% to 0.3% being a well-supported sweet spot for visible benefits with good tolerability.

One quirk worth mentioning: Retinol is famously unstable.

It's highly sensitive to light and oxygen, and UV exposure breaks it down into a range of degradation products.

Real-world testing bears this out, with retinoid content in some products dropping anywhere from 0% to 80% after six months at room temperature, and even more at higher temperatures.

This is why good formulations lean on opaque, air-tight packaging (think tubes and pumps, not clear jars) and often "encapsulate" the Retinol to shield it.

Signs of oxidation include your product turning yellow or smelling "off". Keeping it somewhere cool and dark, and using it up within a few months of opening helps it stay effective.

The most common side effects are mild and temporary: usually some dryness, redness, or light peeling as your skin adjusts. These tend to settle with consistent and lower-frequency use.

Like all retinoids, Retinol works best with nightly use, a good moisturizer, and daytime sunscreen.

The "ramp up" method works well: start with Retinol once a week to give your skin time to adjust, which keeps irritation low. Slowly add more nights until you reach your goal frequency once your skin feels comfortable.

Retinoids also make your skin more sensitive to the sun in the first few weeks, so wear sunscreen every morning and protect your skin from direct sun while you build up tolerance.

One safety note: topical Retinoids aren't recommended during pregnancy or breastfeeding. Systemic absorption from creams is low but because high oral vitamin A is a known teratogen and topical safety data are limited, most clinicians recommend stopping retinoids when pregnant or trying to conceive.

Hyaluronic acid (HA) is a glycosaminoglycan (basically a long sugar chain) that your skin already makes on its own. In your skin, HA lives in the extracellular matrix and acts as the body's moisture reservoir.

Topically, HA is a humectant that binds water and helps skin look more plump, smooth, and hydrated.

The only catch is that HA isn't a single thing; it actually comes in a wide range of molecular weights (~50 - 2,000+ kDA) and size matters.

Some clinical evidence links low molecular weight versions to improved wrinkle depth, elasticity, anti-inflammatory effects, and barrier repair.

This is why the best HA serums blend the two sizes together so you get the best of both worlds.

The majority of cosmetic HA is produced by bacterial fermentation, typically using Streptococcus or Bacillus strains. Typical use levels in skincare sit around 0.1-2%.

A clinical study using a 0.2% low-molecular weight HA gel showed improvement in facial seborrheic dermatitis with excellent tolerance.

3-O-Ethyl Ascorbic Acid is one of the most popular "stable" vitamin C derivatives in skincare.

Plain ascorbic acid is fantastic but notoriously fragile; it browns, oxidizes, and loses potency fast. So attaching an ethyl group to the third carbon of the molecule gives it some cool perks:

In a formula, it does the 3 classic vitamin C jobs: it acts as an antioxidant, helps brighten skin tone by inhibiting tyrosinase, and supports collagen.

The evidence is reasonably solid for a cosmetic ingredient; Liao and colleagues (2018) showed it's significantly more stable than ascorbic acid while still being effective.

A 2021 study by Zerbinati and colleagues tested a serum with 30% 3-O-ethyl-l-ascorbic acid and 1% lactic acid significantly increased collagen production, reduced UVB-induced DNA damage, and decreased melanin on a reconstructed pigmented skin model.

Typical real world usage sits around 0.5-5% (and 1-2% is common for daily serums).

Amounts up to 30% have been shown to be non-irritating on human skin samples, but two isolated cases reported allergic contact dermatitis so a patch test is sensible if you have reactive skin.

Niacinamide is a multitasking form of vitamin B3 that strengthens the skin barrier, reduces pores and dark spots, regulates oil, and improves signs of aging.

And the best part? It's gentle and well-tolerated by most skin types, including sensitive and reactive skin.

You might have heard of "niacin flush", or the reddening of skin that causes itchiness. Niacinamide has not been found to cause this.

In very rare cases, some individuals may not be able to tolerate niacinamide at all or experience an allergic reaction to it.

If you are experiencing flaking, irritation, and dryness with this ingredient, be sure to double check all your products as this ingredient can be found in all categories of skincare.

When incorporating niacinamide into your routine, look out for concentration amounts. Typically, 5% niacinamide provides benefits such as fading dark spots. However, if you have sensitive skin, it is better to begin with a smaller concentration.

When you apply niacinamide to your skin, your body converts it into nicotinamide adenine dinucleotide (NAD). NAD is an essential coenzyme that is already found in your cells as "fuel" and powers countless biological processes.

In your skin, NAD helps repair cell damage, produce new healthy cells, support collagen production, strengthen the skin barrier, and fight environmental stressors (like UV and pollution).

Antioxidant

This ingredient is also known as rucinol. It's an antioxidant and one of the best-studied tyrosinase inhibitors (skin brightener) in cosmetic chemistry.

This ingredient works in a few complimentary ways:

In head-to-head biochemical assays, it outperformed kojic acid, arbutin, and even hydroquinone at slowing melanin production.

Clinically, 0.1% has shown to improve melasma visibly after just 4 weeks. Another liposome-encapsulated version performed similarly well and an Indian study on 0.3% showed significant melasma reduction over 8 weeks with no adverse events reported.

This ingredient is generally well-tolerated. A very small amount of people have experienced mild dryness or peeling at a higher concentration (0.3%).

According to a manufacturer, this ingredient is sensitive to air. Good formulations should account for this to keep your rucinol effective.

Tocopheryl Acetate is a stable, shelf-friendly form of vitamin E.

Formulators love it because plain vitamin E oxidizes quickly once it hits air. This acetate version stays stable and resists going off, helping to extend a product's shelf life.

It's actually inactive on its own and works like a slow-release "storage" form; the enzymes in your skin called esterases gradually convert it into active vitamin E over time.

One in vivo study showed 5% of the acetate in the living layer of the epidermis converted to vitamin E after 5 days of application. This study also found the skin gained protection against UV damage even though the conversion was slow and small.

Once converted, vitamin E acts as a skin's main fat-soluble antioxidant that fights free radicals to protect skin from damage.

Topical vitamin E generally boosts the skin's photoprotection, and it reduced UV-damage in animal models.

This ingredient has some brightening potential but it's more of a prevention ingredient than spot-fader. Cell studies show it can slow down melanin production but it's worth noting that it's not the most powerful brightener out there.

Overall, it has a pretty solid safety profile and has been found to be non-irritating and non-comedogenic. Allergic reactions may happen but stay rare due to how widely the ingredient gets used.

The concentration will vary depending on the formula; industry data shows 0.1% in baby lotions, 3% in lipsticks, and 5% in foot powders. You can also find this ingredient at 100% in a pure vitamin E oil.

Most leave-on skincare keeps it at the lower end, often between 0.5-1%.

Collagen is a big structural protein that your body uses to keep skin firm and bouncy. Despite the marketing, topically applied collagen doesn't "refill" the collagen in your skin.

The molecule is too big to pass through your skin barrier so intact Collagen physically can't get past the surface.

What it actually does in your skincare is work as a humectant and film-former: it binds water, lays down a light moisturizing film, and reduces water loss from the surface. This helps make skin feel smoother and temporarily plumper.

This ingredient has been found safe for use in cosmetics with clinical studies showing no irritation, sensitization, or phototoxicity. It's typically used at low concentrations (often a fraction of a percent up to a few percent).

Collagen will not increase sun sensitivity, but you should always wear sunscreen during the day.

Alpha-Arbutin is a brightening active used to fade dark spots, melasma, and post-acne marks. Structurally, it is a hydroquinone with a sugar molecule attached (but is much gentler than raw hydroquinone).

It has a pretty elegant way of working; it slots into tyrosinase, or the key enzyme that your skin uses to make melanin), and competitively blocks it so your skin produces less pigment over time.

There are two forms of arbutin and alpha is the better one to reach for: it works at lower amounts, absorbs into skin more easily, and has a strong safety record.

The science backs this up too: lab and animal studies confirm it lowers melanin by blocking tyosinase and the more you use (up to a point), the more it works.

Human studies look good too.

A 2024 trial found a cream with 5% alpha-arbutin and 2% kojic acid worked about as well as a prescription melasma cream but with fewer side effects. Another study showed 2% alpha-arbutin paired with 10% Trihydroxybenzoic Acid Glucoside and sunscreen faded dark spots and melasma without causing irritation.

Concentration-wise, most serums land in the 1-2% range.

The EU's Scientific Committee on Consumer Safety concluded that alpha-arbutin is safe in face creams up to 2% and body lotions up to 0.5%.

Overall, this is a well-studied and low-irritation brightener that's a great pick for anyone dealing with uneven skin tone. It also pairs well with vitamin C, niacinamide, and sunscreen.

Glutathione is a tiny protein-like molecule (a "tripeptide" build from 3 amino acids: glycine, cysteine, and glutamic acid) that your body already makes on its own.

Inside your body, it acts as one of the skin's main antioxidants that help fight against free radicals.

In skincare, it's best known as a brightening ingredient that slows down tyrosinase, the key enzyme that makes skin pigment. It also nudges the skin toward making a lighter type of pigment instead of a darker one.

This is why you'll see it in products aimed at dark spots and uneven tone.

A small number of real human trials have found a topical glutathione lotion:

The honest caveat is that the current evidence is still thin (few studies, small groups, short timelines). Glutathione also doesn't absorb into skin very easily so results tend to be modest and fade if you stop using it.

One thing worth clearing up:

The scary side effects you may have heard about come from glutathione injected intravenously, which has real safety concerns. Applying it topically is a completely different thing and has a clean track record.

Most human studies used it around 2% (as Glutathione or Glutathione Disulfide) and a 2% oxidized glutathione lotion and a 2% S-acyl glutathione cream are the concentrations with actual clinical data behind them.

There's no established "ideal" percentage yet but 1-2% is the evidence-backed range.

Allergy-wise, there is very low risk for this ingredient; it was well-tolerated across the topical trials. Only one participant had mild temporary redness that cleared up on its own and another study reported no adverse reactions at all.

One trial had ~10% of users drop out for irritation was using a combination cream that also had 10% azelaic acid so the irritation likely wasn't from the glutathione. There's no notable contact-allergy signal for topical glutathione in the literature but patch-testing before first use is still sensible for those with sensitive skin.

Camellia Sinensis Leaf Extract (tea extract) is one of the most well-researched plant extracts in skincare with an impressive resume.

Black tea, green tea, and oolong tea are all harvested from the Camellia Sinensis plant.

Studies show green tea extract and its catechins (like epigallocatechin gallate (EGCG)) help your skin cells product energy more efficiently and reducing the number of free-radicals that can damage your skin from the inside.

In lab-grown skin models, this translated to younger, healthier, and stronger skin.

There's also good sun protection data; researchers saw less DNA damage and redness on human skin when green tea was applied before UVB exposure. And the more they applied, the better the protection.

Needless to say, this ingredient shouldn't replace your sunscreen. But it is a great supportive ingredient that you can already find in many sunscreens and antioxidant serums.

A 2009 study found a 2% green tea lotion was effective for mild-to-moderate acne thanks to its anti-inflammatory and mild antimicrobial activity.

The quality of the extract matters a lot here:

Good extracts contain 50-90% catechins while lower quality ones are mostly there for marketing. We recommend reaching out to the brand if you have questions about the quality or source of their ingredients.

Human Repeated Insult Patch Testing showed no irritation or sensitization at use concentrations (0.86% in leave-on products and up to 30% as leaf water).

Dipotassium Glycyrrhizate comes from licorice root.

Extracts of licorice have demonstrated to have antibacterial, anti‐inflammatory, antiviral, antioxidant properties.

One component, glabridin, has extra potent antioxidant and soothing properties. It has also been found to block pigmentation from UVB rays in guinea pigs.

Licorice Root also contains a flavonoid. Flavonoids are a natural substance from in plants. Flavonoids also have antioxidant properties.

Another component, glycyrrhizin, has been found to have anti-inflammatory and antimicrobial benefits. This may make licorice root extract effective at treating acne. However, more research is needed to support this.

Liquiritin is one of the flavone compounds found in licorice. It has been found to help lighten skin by preventing tyrosinase from reacting with tyrosine. When the two react, protein is converted to melanin. Melanin is the substance in your body that gives your features pigmentation.

Licorice root is native to Southern Europe and Asia. It has been used in traditional Chinese medicine to help with respiratory issues.

Bisabolol is a gentle skin conditioner, antioxidant, and soothing ingredient.

It's primary claim to fame is soothing and research shows topically applied bisabolol can quiet the chemical messengers that cause your skin to become inflamed, helping to sooth any irritation.

A clinical study found that applying 0.5% bisabolol daily for 8 weeks produced an average 9% decrease in skin pigmentation. Researchers found it can also suppress the process that leads to excess melanin production in skin.

In vitro studies found that bisabolol combined with propylene glycol significantly increased skin permeability by increasing lipid fluidity in the stratum corneum.

You'll likely see use concentrations quite low, usually 0.1-0.2%.

Overall, this is a well-tolerated ingredient that works well in formulas designed for sensitive, reactive, or post-procedure skin.

You may know this ingredient as argan oil. It has emollient and skin conditioning properties that help soften skin and reinforce the lipid barrier.

The fatty acid profile of argan oil is roughly 45-55% oleic acid, 28-36% linoleic acid, 10-15% palmitic acid, and 5-7% stearic acid. It also contains vitamin E, sterols, squalene, and polyphenols like ferulic acid.

Two clinical studies in postmenopausal women found that applying argan oil for 60 days significantly improved skin elasticity and moisturization (reduced transepidermal water loss and increased epidermal water content).

Since it is high in oleic and linoleic acid, this ingredient may not be fungal acne safe. Both of these fall in the C11-C24 range that Malassezia yeast can metabolize.

Bakuchiol is a plant-derived antioxidant from the seeds of the Psoralea corylifolia plant. It has antimicrobial, emollient, skin conditioning, and antioxidant properties.

You'll likely see it called a "retinol replacement" but the two are technically not related. This is because bakuchiol is able to flip many of the same switches in your skin cells to tell them to:

1) produce more collagen (type I, III, and IV)
2) activate the same genes retinoids do

Unlike retinoids, this ingredient will not increase photosensitivity and is safe to use during pregnancy (but please still check in with your doctor!).

The flagship clinical trial from Dhaliwal et al. 2019 found 0.5% bakuchiol (twice daily) and 0.5% retinol (once daily) reduced wrinkles and hyperpigmentation equally, but bakuchiol had significantly less irritation.

Systematic reviews also back this up:

Bakuchiol is comparable to retinol for photoaging but with better tolerability. It also has mild antibacterial properties against Cutibacterium acnes and antifungal activity in vitro against Candida and dermatophytes.

The reason bakuchiol works well is due to its structure; it is a meroterpene phenol, or a hybrid molecule. The phenol half acts as an antioxidant while the terpene half is fat-loving. This helps the molecule slip through the skin barrier.

This ingredient is usually used between 0.5-2%. Only one case of contact dermatitis has ever been reported for this ingredient.

Acetyl Hexapeptide-8 (also known as Argireline) is a synthetic hexapeptide that is often called a "topical Botox alternative".

It works by mimicking how Botox relaxes muscles; it interferes with the signaling process that tells your facial muscles to contract. This can help soften expression lines like forehead wrinkles or crow's feet over time.

The comparison to Botox does have limits because the molecule is water-loving and relatively large.

Acetyl Hexapeptide-8 has a hard time absorbing deeply enough through the skin's outer barrier to actually reach the muscles.

So whether it truly works the way Botox does at a biological level is still up for debate, but early clinical outcomes are fairly encouraging.

While some studies have observed improvements in wrinkle appearance, it is important to note that more consistent results are seen in multi-ingredient formulations (vs just Acetyl Hexapeptide-8 alone).

Some research studies also used higher concentrations (up to 10%) while this ingredient is usually found in concentrations up to 0.005% in leave-on formulations.

Hexapeptide-2 is a peptide.

Cosmetic Colorant, Skin Protecting, UV Absorber

Zinc Oxide (ZO) is a mineral broad-spectrum UV filter and the broadest-spectrum filter recognized by the FDA. It covers everything from UVB through to long-wave UVA.

On top of sun protection, it has skin protectant and skin-soothing properties too.

Here's a myth worth busting: mineral filters are usually described as working by "reflecting" or "bouncing" UV off your skin.

That's mostly not true: when researchers actually measured it, ZO and Titanium Dioxide reflect only about 4-5% of UV (less than SPF 2 worth of protection).

The vast majority of the work (~95%) is done by absorption, similar to chemical UV filters. ZO is a semiconductor that absorbs UV photos through its energy band gap.

So the old "physical blocker vs. chemical absorber" framing is really an oversimplification.

Zinc Oxide is one of the most effective broad-spectrum UV filters out there. It protects across UVB, UVA2, and UVA1 with a flat, even absorption curve across the whole UVA-UVB range.

That uniform UVA coverage is its standout feature; titanium dioxide skews more toward UVB as its particle size drops so ZO gives more consistent and extended UVA protection.

It's also very photostable. As an inorganic oxide, ZO doesn't break down in sunlight the way some organic filters can, so it holds up over a day of wear.

This ingredient is gentle and soothing, making it go-to for sunscreens aimed at sensitive skin, rosacea, or ecezma-prone skin, babies, and children.

It's also unlikely to cause the "eye sting" that some sunscreen ingredients are known for, and regulatory agencies broadly consider it non-toxic and safe for topical use.

Beyond sun protection, ZO is also a recognized OTC skin protectant. It forms a breathable barrier that shields skin from moisture and irritation while supporting healing. This is why you'll see it as a classic active in diaper rash creams.

The only downside to ZO is that it can leave a visible white cast, especially on deeper skin tones. This is the main reason mineral sunscreens have historically felt less cosmetically elegant than chemical or hybrid formulas.

Zinc Oxide comes in both non-nano and nano forms. The dividing line is 100nm and anything under is classified as a nanomaterial by the EU.

The nano version scatters less visible light which cuts down white case and gives a lighter, more wearable texture.

Another thing worth understanding about formulation:

Uncoated ZO has some inherent photocatalytic activity. This just means it can generate reactive oxygen species under UV. It's exactly why cosmetic-grade ZO is almost always surface-coated; this coating suppresses that reactivity and improves how the powder disperses and feels.

A well-formulated coated ZO largely sidesteps this issue.

Zinc Oxide is commonly used anywhere from 10% up to the regulatory maximum in sunscreens (25%).

Mineral-only broad-spectrum products often land in the 15-25% range to hit higher SPF and UVA values. Keep in mind SPF performance depends heavily on particle size, dispersion, and the rest of the formula, and not just the percentage.

As an OTC skin protectant like diaper creams, ZO typically runs higher at roughly 10-40%.

This ingredient is generally easy to work with and doesn't photodegrade.

The only thing to know is that uncoated ZO can be a bit reactive in a formula.

Under UV, it can break down sensitive ingredients like other actives or UV filters. This is another reason coated versions are standard. ZO can also react with very acidic ingredients or throw off stability of some creams. A good formula will get around this with the right coatings and dispersion.

The EU's Scientific Committee on Consumer Safety has concluded that ZO nanoparticles "can be considered to not pose any risk of adverse effects in humans after application on healthy, intact or sunburnt skin".

You might hear that ZO is "toxic"; this is because an in-vitro (test tube) study suggested micronized ZO had potential phototoxicity. In vivo (human) investigations have disputed this and the results have come back reassuring.

So does ZO penetrate skin? The short answer is no, not in any way that matters.

The most relevant evidence comes from real-world human studies: in one, volunteers applied ZO nanoparticle sunscreen hourly for six hours and daily for five days. The advanced imaging showed the particles stayed on the surface and never reached the living epidermis, and no cellular toxicity was found.

A small amount of solubilized zinc ions can dissolve off the particles and enter the upper skin. But the quantities are tiny compared to the zinc already naturally present in your body, and studies haven't found this to cause local toxicity.

The sunscreen bans you've heard of (like Hawaii's) are aimed at two chemical filters, Oxybenzone and Octinoxate. ZO itself it not banned and is often recommended instead.

TD gives broad-spectrum protection that's strongest in the UVB and UVA-2 range and weaker in the UVA-1 range. Its UVA protection isn't quite as strong as Zinc Oxide's which is why you'll often see the two paired together.

There's also no evidence of carcinogenicity, mutagenicity, or reproductive toxicity from dermal exposure of this ingredient.

For those who have seen the headline about a 2022 EU ban on TD, that was on TD as a food additive (a complete separate use from topical sunscreen).

There are ongoing questions about how nano-TD might affect marine ecosystems. As of now, there has been no conclusive evidence that any form of TD (or any other sunscreen filter) harms coral reefs or marine life.

The science is still developing and it's a space worth watching rather than packing over.

However, several destinations have reef-safety sunscreen rules that restrict certain chemical filters and steer visitors toward mineral, non-nano options. If you're traveling somewhere with these rules, a non-nano mineral sunscreen is the safe bet.
